摘要 目的:探讨咪唑安定对谷氨酸诱导大鼠神经元样PC12细胞凋亡的影响。方法:诱导分化4 d的神经元样PC12细胞随机分成空白对照(C组),加入20 mM谷氨酸(G组),加入20 mM谷氨酸和10 uM咪唑安定(M组),各组细胞继续培养24 h后观察细胞的活力(MTT法)及细胞凋亡率(Hoechst33258核染色法联合Annexin V-FITC/PI双染流式细胞术)。结果:与C组比较,G组细胞活力度明显降低,而细胞凋亡率明显升高(P<0.01);与G组比较,M组细胞活力度明显升高,而细胞凋亡率明显降低(P<0.05)。结论:咪唑安定可抑制谷氨酸诱导体外培养的大鼠神经元样PC12细胞凋亡,发挥保护作用。 Objective: To explore the effects of Midazolam on glutamate-induced apoptosis in neuronal PC12 cells. Methods: The differentiated neuronal PC12 cells were randomly allocated to be exposed to new culture medium F(group C) or glutamate 20mM (group G) or glutamate 20 mM add Midazolam 10 uM (group M) and incubated for 24 hours. Then, MTT assay was applied to evaluate the viability of cells, hoechst33258 nucleus staining and flow cytometry with Annexin V-FITC/PI staining were performed to detect the apoptosis. Results: The viability of PC 12 cells was decreased and the apoptosis increased significantly in group G compared with group C(P < 0.01). The cell viability increased and the apoptosis ratios decreased significantly in group M compared with group G (P< 0.05). Conclusion: Midazolam protects neuronal PC12 cells from glutamate-induced apoptosis.
