摘要
目的:探讨抑癌基因PTEN及凋亡抑制蛋白Livin蛋白在舌癌(Tongue Squamous Cell Carcinoma TSCC)中表达水平,与临床病理指标的关系及预后意义。方法:免疫组织化学S-P方法检测10例正常舌黏膜和45例舌癌标本中PTEN及Livin蛋白的表达,并与组织学类型、组织学分级、淋巴转移和临床分期等生物学行为进行相关分析。结果:1PTEN主要表达在细胞浆中。正常舌黏膜中全部阳性表达,TSCC组织中阳性表达率为45.0%,2Livin主要表达在细胞浆中。正常舌黏膜中全部阴性表达,TSCC组织中Livin蛋白表达率为53.0%,两者阳性表达与舌癌高、中、低分化;有、无淋巴结转移相关(P<0.05),与舌癌患者的性别、年龄、肿瘤大小无明显相关性(P>0.05)。结论:PTEN和Livin的表达与舌癌发生、发展密切相关,在TSCC组织中PTEN蛋白的阳性表达率较低,说明PTEN失活在TSCC的发生发展中起着一定的作用,Livin抑制TNF-a和细胞色素C通路,表现为抗细胞凋亡,二者的联合检测有助于口腔鳞癌的早期诊断及判断预后指标。
Objective:To investigate the expression of the tumor suppressor gene PTEN and protein inhibitor of apoptosis protein Livin in Tongue Squamous Cell Carcinoma(TSCC) and its relationship with clinicopathological parameters and prognostic significance.Methods:We detected the expressions of PTEN and Livin in 45 cases of TSCC and 10 cases of normal tongue tissue by using immunohistochemistrical SP method,and analyzed the relationship between the expressions and the biological behavior,such as histological type,histological grade,lymph node metastasis and clinical staging.Results:①The PTEN expressions was in the cytoplasm.In the normal tongue mucosa,it showed all positive expression,and in TSCC,the ratio of positive expression was 45.0%,②The Livin expressions was in the cytoplasm.In the normal tongue mucosa,it showed all negative expression,and in TSCC,the ratio of positive expression was 53.0%.Both of their expression were correlated with histological grade,and lymph node metastasis(P<0.05),but had no correlation with patients′sex,age and tumor size(P>0.05).Conclusion:The expression of PTEN and Livin is closely related to the occurrence and development of tongue cancer.In TSCC,the ratio of PTEN positive expression is less,suggesting the PTEN inactivation plays a certain role in the occurrence and development of TSCC.Livin inhibits TNF-a and cytochrome C pathway,manifesting as anti-apoptosis.Both of the their detection can help earlier diagnosis of TSCC and determine prognostic indicators.
