摘要
目的探讨基质金属蛋白酶2(matrix metalloproteinase 2,MMP-2)及组织金属蛋白酶抑制剂2(tissueinhibitor of metalloproteinase 2,TIMP-2)在人肺腺癌中的表达及其意义。方法应用链霉素抗生物素蛋白过氧化物酶法染色技术,检测60例肺腺癌及30例癌旁正常肺组织中MMP-2和TIMP-2的蛋白表达。结果肺腺癌与正常肺组织中,MMP-2阳性表达率分别为73.3%和13.3%,TIMP-2阳性表达率分别为46.7%和10.0%,差异均有统计学意义(P<0.05);肺腺癌的不同分化程度MMP-2的阳性表达与不同TNM分期MMP-2的阳性表达差异均有统计学意义(P<0.05);生存期<2年患者的MMP-2阳性表达率明显高于生存期≥2年患者(P<0.05);与无淋巴结转移的MMP-2阳性表达差异有统计学意义(P<0.05)。结论 MMP-2阳性表达上调,MMP-2/TIMP-2失平衡,MMP-2蛋白过剩,可能是导致肺腺癌发生的主要原因之一;可作为肺腺癌恶性程度及预后判断的指标。
Objective To investigate the expression of matrix metalloproteinase 2 (MMP-2) and tissue inhibitor of metalloproteinase 2 (TIMP-2) and their correlation with metastasis and prognosis in human lung adenocarcinoma.Methods Immunohistochemical method was used to detect the expression of MMP-2 and TIMP-2 in 60 cases of lung adenocarcinoma tissues and 30 cases of lung paracancerous tissues.Results The positive expression rates of MMP-2 in lung adenocarcinoma tissue and paracancerous tissue were 73.3% and 13.3% respectively,whereas the positive rates of TIMP-2 expression in lung adenocarcinoma tissue and paracancerous tissue were 46.7% and 10.0% respectively.Significant difference of the expression rates of MMP-2 and TIMP-2 were found between lung adenocarcinoma and paracancerous tissue ( P <0.05).The positive expression of MMP-2 showed statistically significant differences in different differentiation of lung adenocarcinoma ( P <0.05).The positive expression of MMP-2 showed statistically significant differences in different TNM stage of lung adenocarcinoma ( P <0.05). Between the survival time <2 years group and ≥2 years groups,the expression of MMP-2 had significant difference (P < 0.05).The positive expression of MMP-2 showed statistically significant differences between with metastasis and without metastasis ( P <0.05 ). Conclusion Overexpression of MMP-2 and the imbalance of MMP-2 and TIMP-2 are major reasons for the lung adenocarcinoma carcinogenesis.It can act as a reference index for the malignant level of lung adenocarcinoma and prognosis.
