细胞凋亡相关因子Caspase-3、Caspase-9和增殖细胞核抗原在人肾透明细胞癌中的表达 被引量:2

摘要 目的检测人肾透明细胞癌组织中细胞凋亡相关重要因子Caspase-3、Caspase-9和增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)表达的变化,研究这些因子的表达与肾癌的关系。方法标本离体后分成3组,①肾癌细胞组;②癌旁组织组;③正常肾组织组。均用10%福尔马林固定48h,常规石蜡包埋、切片5μm,应用免疫组织化学方法进行观察。结果 PCNA免疫组织化学染色结果,正常肾组织内的细胞核有弱的阳性表达,癌旁组织阳性表达增加,高于正常肾组织;肾癌组织阳性表达明显增强,明显高于癌旁组织。Caspase-3与Caspase-9均在正常肾组织中有弱的阳性反应,癌旁组织阳性表达增加,肾癌组织表达明显减少。3组之间两两比较差异均有统计学意义(P<0.01)。肾癌4个期之间比较差异无统计学意义(P>0.05)。结论癌旁组织Caspase-3和Caspase-9蛋白表达明显高于正常肾组织及癌组织,癌组织表达最少;癌组织中Caspase-3和Caspase-9表达下降,说明细胞凋亡减少;PCNA在正常肾组织组、癌旁组织组、肾癌组织组的表达呈明显加强趋势,癌组织细胞增殖旺盛,是肿瘤形成的重要机制。Caspase-3、Caspase-9和PCNA于肾透明细胞癌的表达在肿瘤分期间差异无统计学意义。Caspase-3与Caspase-9蛋白表达呈正相关。 Objective The study was aimed to detect the expression of apoptosis-related factors Caspase-3,Caspase-9 and proliferating cell nuclear antigen (PCNA) in human renal clear cell carcinoma (RCCC) tissues and to explore the correlations of these factors with RCCC. Methods Samples were divided into three groups,RCCC group,para-cancerous tissue group,and normal tissue group.All samples were fixed in formalin for 48 h,embedded in paraffin wax,cut into sections at 5μm thickness,and studied by immunohistochemical method.Results The expression of Caspase-3 and Caspase-9 were weakly positive in the normal renal tissue,increased in the para-cancerous tissue and significantly decreased in the RCCC tissue.The expression of PCNA was weakly positive in the nucleus of the normal renal tissue,slightly higher in the para- cancerous tissue and the highest in the RCCC tissues among the three tissues.There were significant differences among the three groups in each factor ( P <0.01),but no significant difference among 4 stages of renal carcinoma (P > 0.05).Conclusion The expressions of Caspase-3 and Caspase-9 protein in para-cancerous tissue were significantly higher than normal renal tissue,and the lowest in RCCC tissue.The results suggested that cell apoptosis reduced in RCCC.The expression of PCNA in RCCC tissue was higher than para-cancerous tissue and normal renal tissue.The increased cell proliferation were one of the important mechanisms in the tumor formation.There were no significant differences of Caspase-3,Caspase-9 and PCNA expressions among different stages of renal clear cell carcinoma.
