摘要
建立一种苦瓜多糖含量测定方法。苦瓜样采用80%乙醇回流除去单糖、双糖、低聚糖、多酚、氨基酸等杂质后,采用热水回流提取的方法从苦瓜样品中分离提取苦瓜多糖。为了减少测定误差,采用精制苦瓜多糖对葡萄糖的换算因子,以葡萄糖标准液绘制标准曲线,用蒽酮-硫酸分光光度法对苦瓜样品中的苦瓜多糖含量进行测定,检测波长为625nm。实验结果显示葡萄糖标准曲线的回归方程为Y=6.965×C-0.037(R2=0.9974),线性范围在0~200μg之间,表明该测定方法简便可行,供试液在4h内显色稳定,重现性较好,平均回收率为100.58%,RSD为1.21%(n=3),对苦瓜中多糖含量的测定可获得满意的结果。实验结果表明蒽酮-硫酸法操作简便,重现性好,准确,可靠,可作为检测苦瓜多糖含量的优选方法。
