摘要
利用在PDA培养基中添加愈创木酚和α-萘酚的平板法,对43株大型真菌进行产漆酶初筛,根据平板菌落周围产生的褐色和紫色氧化圈的显著程度初步筛选得到明显分泌漆酶的21个菌株.进一步将这21个菌株分别进行摇瓶发酵,用ABTS法检测发酵液中漆酶的活力,复筛出3个漆酶产量较高的菌株,分别为香菇L03、灵芝T1和糙皮侧耳5.33,其酶活分别为382.3、324.9和297.1 U.L-1.
Through primary screening methods on guaiacol-and α-naphthol-containing plates,21 fungi strains which produced laccase obviously were screened from 43 macro-fungi strains according to the diameters and coloration of oxidative circle resulted from reaction catalyzed by the enzyme.By analyzing laccase activity of 21 strains in liquid medium with ABTS as substrate,three high-efficient laccase-producing strains were obtained.These strains are Lentinula edode L03,Ganoderma lucidum T1 and Pleurotus ostreatus 5.33...
