摘要
采用铝箔纸包裹的方法,对生长中的烟草单个叶片进行黑暗处理(DIS)。发现在处理第5d叶片明显变黄,叶片衰老标记基因CP1表达量开始明显增高;与黑暗诱导的离体叶片相比,黑暗诱导的生长叶片在叶绿素、可溶性蛋白、MDA含量以及SOD、POD酶活等指标上,呈现相似但较为平缓的变化趋势。该结果可为进行烟草叶片衰老研究提供理想的实验体系。
Individual tobacco leaf was kept in dark environment in vivo by aluminum foil paper to induce senescence and to study the process of quality formation.RNAs of senescence-specific cysteine protease(CP1)were detected wiht RT-PCR on 3rd day after treatment,and leaf became yellowish on 5th day.A comparative analysis of darken individual leaf in vivo(DIS)and dark-incubated leaf in vitro(DET)revealed similar changes in chlorophyll,soluble protein content and SOD,POD activity during darkening.Both senescence proce...
