摘要
目的探讨组蛋白甲基转移酶Ezh2在P19神经细胞分化中的作用。方法通过Western印迹检测全反式维甲酸(RA)诱导TuJ1蛋白的表达;通过染色质免疫沉淀技术检测Ngn1基因启动子区Ezh2的结合与H3K27三甲基化的变化;通过实时定量RT-PCR检测RA诱导P19细胞分化后Ezh2mRNA表达。结果在RA诱导P19细胞中,Ezh2与H3K27me3在Ngn1启动子区的结合逐渐升高,并在诱导后第2天达到高峰,然后迅速降低,随后出现神经分化特异标志。结论Ezh2在RA诱导的P19细胞早期产生一种抑制性的组蛋白修饰标志H3K27me3,通过避免Ngn1的过早表达,确保P19细胞的正常分化。
