大黄总蒽醌含药血清对LoVo的AQP4表达的调节效应

Regulating effects of aquaporin4 expression in LoVo cells by serum containing total anthraquinone in rheum

目的:探讨大黄总蒽醌含药血清对体外培养LoVo细胞的水通道蛋白基因(AQP4)表达的调节效应。方法:16只SD大鼠随机分为正常组,大黄总蒽醌低、中、高剂量组(1.4,2.5,4.5 g.kg-1.d-1,灌胃),7 d后麻醉处死取血制备含药血清。体外培养LoVo细胞并给予不同剂量大黄总蒽醌含药血清培养液24 h,采用W estern b lot及半定量RT-PCR检测AQP4蛋白及mRNA的表达。结果:结果显示,4.5 g.kg-1.d-1大黄总蒽醌含药血清培养液可抑制LoVo细胞的AQP4蛋白及mRNA的表达(P<0.01)。结论:大黄总蒽醌含药血清可抑制LoVo细胞AQP4基因转录与翻译,提示大黄的泻下作用可能与调节AQP4表达有关。
