摘要 目的 观察重组人生长激素对大鼠肝硬化肝细胞代谢活性的影响。方法 采用胶原酶原位灌注游离大鼠肝硬化肝细胞,以重组人生长激素(13.3、133.3、1333ng/ml)培养36 h,检测其干预效果。结果 培养后肝硬化肝细胞浓度无显著性变化(P>0.05),上清液中的白蛋白含量增加(P<0.05),乳酸脱氢酶活性降低(P<0.05),肝硬化肝细胞的甲基偶氮唑盐显色活性增加(P<0.05)。结论 重组人生长激素对离体大鼠肝硬化肝细胞的代谢活性有明显的刺激作用。 Objective To investigate the effect of recombinant human growth hormone on the metabolic activites of liver celis in vitro of cirrhotic rats. Methods Liver celis were isolated through collagenase perfusion in situ of rat livers with cirrhosis and cultured with recombinant human growth hormone (13. 3ng 133. 3ng 1333ng/ ml) for 36 hours. Results After the culture, the concentration of liver celis did not change significantly ( P > 0. 05), but the quantity of albumin in the supernatant was increased ( P < 0. 05), the activity of lactic dehydroge-nase (LDH) was decreased ( P < 0. 05) and the cellular activity of tetrazolium (MTT) bio-reduction was increased ( P < 0. 05) significantly. Conclusions The metabolic activities of liver celis in vitro of cirrhotic rats have an obvious stimulation by recombinant human growth hormone.
