摘要
目的 探讨生物人工肝对重型肝炎的治疗效果。方法 通过猪肝细胞的分离与培养,建立猪肝细胞型生物反应器,与日本KM8800型血浆交换仪共同构建生物人工肝体外支持系统,在体外研究了猪肝细胞型生物人工肝对重型肝炎血浆生化指标及药物转化代谢的影响。结果 该型生物人工肝对重型肝炎患者血浆中的总胆红素、谷氨酰转肽酶、胆碱酯酶、血糖、胆固醇及糖化血清蛋白的水平有所下降,对总蛋白、白蛋白、甘油三脂水平增加,氨茶碱浓度迅速下降。结论 猪肝细胞型生物人工肝系统能参与糖、蛋白、脂肪的代谢,有较好的生物合成和生物转化功能,对重型肝炎有一定的肝功能支持作用。
Objective To study the effect of bioartificial liver support system on the patient with severe hepatitis. Methods The bioartificial liver support system was composed of the hollow fiber bioreactor and the KM8800 plasma exchange device. The cultured porcine hepatocytes in the bioreactor were isolated from the young pig with about weight of 15kg. The biologic function of bioartificial liver on plasma biochemistry of patients with severe hepatitis and on drug biotransformation were observed by applying the device circulating in vitro. Results The levels of total serum bilirubin, r-glutamyl transpeptidase, cholinesterase, glucose, cholesterol and serum glycoprotein in the plasma from the patient with severe hepatitis were lowered and the levels of total protein, albumin, triglycerides were elevated after the treatment of bioartificial liver by circulating in vitro. The content of aminophylline in plasma was decreased rapidly after the treatment as well. Conclusion The bioartificial liver with porcine hepatocytes can participate the metabolism of sugar, lipids and protein and has both the biosynthesis and biotransfomation functions. It is useful in liver function support to the patient with severe hepatitis.
