微波酸水解提取及HPLC法测定葛根中的活性成份葛根素 被引量:3

摘要 研究了微波酸水解法提取葛根中的葛根素(puerarin),并将所拟提取方法同超声酸水解、微波辅助萃取、冷凝回流及室温冷浸4种方法进行对比。通过单因素实验及正交实验,考察盐酸浓度、乙醇浓度、反应温度和反应时间对葛根素提取率的影响。用葛根素测定线性回归方程为Y=2.22×107ρ-456131.5(r=0.9994),加标回收率为96.3%~103.6%,RSD为3.2%。方法可用于葛根药材及葛根素类产品的质量检测。 An acidified microwave-assisted extraction combined with HPLC-UV analysis method for extraction and determination of puerarin in Puerariae radix has been established.Concentration of HCl,concentration of ethanol,extraction time and reaction temperature were optimized by single factor test.A Diamonsil C18 column (150 mm×4.6 mm,i.d.,5 μm,was used,and methanol-0.4% acetic acid solution (40:60,v/v) was used as mobile phase with a flow rate of 0.80 ml min-1.The detection wavelength was 254 nm.The column temperature and sample volume were 25 ℃ ande 10 μL,respectively.The calibration curve for puerarin was linear with the correlation coefficients of 0.9996.The mean recovery of puerarin was 100.2%,and the RSD was 3.2%.The results indicated that acidified microwave-assisted extraction might provide a rapid method for the quality control of Puerariae Radix.
