大潮气量机械通气对大鼠肺组织白介素-1β表达的影响 被引量:1

Effect of high tidal volume mechanical ventilation on the expression of interleukin-1β in the pulmonary tissue of rat

摘要 目的研究大潮气量机械通气对大鼠肺组织白介素-1β(IL-1β)表达的影响。方法健康SD大鼠24只随机分为3组,对照组(A组)不行机械通气,常规通气组(B组)VT为8ml/kg,大潮气量通气组(C组)VT为40ml/kg。机械通气2h后处死动物,采用考马斯亮蓝染色法测定支气管肺泡灌洗液(BALF)中总蛋白的水平,光镜下行白细胞计数,同时测定肺湿干重比值(W/D)和中性粒细胞髓过氧化物酶(MPO)活性;采用ELISA法检测IL-1β水平,RT-PCR法检测IL-1βmRNA含量。结果通气2h后,与对照组相比,大潮气量通气组BALF中总蛋白、白细胞计数、肺W/D、MPO、IL-1β和IL-1βmRNA表达均显著增加(P<0.05或P<0.01),而B组上述各项指标的变化均无统计学意义(P>0.05)。结论大潮气量机械通气可引起大鼠急性肺损伤,诱导IL-1β表达增加。 Objective To examine the effect of high tidal volume mechanical ventilation on the expression of interleukin-1β(IL-1β) in the pulmonary tissue of rat.Methods 24 healthy sprague dawley(SD) rats were randomly divided into control group,conventional ventilation group(VT=8ml/kg) and ventilator associated lung injury group(VALI, VT=40ml/kg).Rats were sacrificed and the lung lavage liquid and lung tissue were collected and stored with correct methods. The contents of total protein,white blood cell(WBC),MPO and ratio of wet/dry lung tissue(W/D) were measured.The level of IL-1βprotein and IL-1βmRNA was dertermined by ELISA and RT-PCR respectively.Results The number of WBC and the value of MPO in bronchial alveolar lavage fluid(BALF),the ratio of W/D,the levels of total protein of IL-1βprotein and IL-1βmRNA in BALF significantly increased in VALI group(P<0.05) compared with conventional ventilation group or control group.Conclusion High tidal volume ventilation induces the expression of IL-1βand causes lung injury.
