摘要
目的通过对3387名大连汉族人群人类白细胞抗原(HLA)-DRB1位点进行分型,扩充了大连地区人群的HLA分型数据库,为群体遗传学提供数据积累。方法采用SSO分型试剂盒对HLA-DRB1进行分型,Luminex 100 IS分析系统全自动结果判读和基因分析,所得数据进行统计学分析。结果大连汉族3387人的HLA-DRB1位点共检测到HLA-DRB1等位基因13种,基因型90种。HLA-DRB1最常见的等位基因为HLA-DRB1*15,占16.33%,最常见的基因型为09/15。大连汉族HLA-DRB1等位基因频率与北方汉族比较无显著性差异,与南方汉族、欧洲人、日本人、喀麦隆人之间的比较都具有显著性差异。结论大连地区汉族人群的HLA-DRB1等位基因频率有其独特的分布方式,HLA有地区差异。
Objective Polymorphism of 3387 cases of unrelated Han ethnic of Dalian was investigated to develop the human leukocgte antigen(HLA) database of Dalian to contribute to anthropology research.Methods Sequence specific oligonucleotide probes(PCR-SSO) were used and Luminex 100 IS was used to read the data automatically and analyze and conclude the characteristics of DRB1.Results 13 cases of alleles and 90 cases of genotypes were detected in 3387 unrelated Han ethnic of Dalian,the most common allele is HLA- DRB1*15(16.33%),the most common genotype is 09/15.The allele frequency showed no significant difference compared with Nothern Han of China,but the allele frequency showed significant differences compared with Southern Han of China,European,Japanese and Cameroon.Conclusion The allele frequence distribution of Han ethnic of Dalian presented unique character,and the HLA showed area differences.
