肝癌衍生生长因子与膀胱癌转移的相关性及其对细胞侵袭能力影响 被引量:4

The relationship of hepatoma-derived growth factor with metastasis of bladder carcinoma and its influence on invasiveness of T24 cells

摘要 目的探讨肝癌衍生生长因子(HDGF)基因与膀胱癌转移的相关性,及其稳定表达对膀胱癌细胞侵袭能力的影响。方法应用荧光定量PCR检测原发未转移膀胱癌和原发伴转移膀胱癌组织中HDGF mRNA的表达。设计并合成HDGF特异性的siRNA,转染膀胱癌T24细胞系,检测转染后细胞侵袭能力的变化。结果原发伴转移膀胱癌组织中HDGF mRNA表达较原发未转移膀胱癌中明显上调(P<0.05)。RNA干扰后,HDGF蛋白明显降低,细胞侵袭能力也随之明显降低(P<0.05)。结论 HDGF基因与膀胱癌的转移相关,其表达下调能够抑制T24细胞的侵袭能力。 Objective To study the relationship of hepatoma-derived growth factor(HDGF) with metastasis of bladder carcinoma,and the influence of HDGF on invasiveness of T24 cell line.Methods Fluorescent quantitation PCR was used to detect the HDGFmRNA in primary bladder carcinoma without metastasis and primary bladder carcinoma with metastasis.The specific siRNA for HDGF was constructed and transfected into T24 cells,then invasiveness change of T24 cell line was detected after transfection.Results The expression of HDGF mRNA was higher significantly(P<0.05) in primary bladder carcinoma with metastasis than in primary bladder carcinoma without metastasis.The expression of HDGF protein decreased significantly in transfected T24 cells,and the invasiveness ability reduced subsequently(P<0.05). Conclusions HDGF gene is positively related to the metastasis of bladder carcinoma,downregulation of HDGF can inhibit the invasiveness of T24 cell line.
