摘要
目的:观察糜酶对自发性高血压大鼠(SHR)心肌纤维化的影响及作用机制。方法:采用RT-PCR检测心肌组织Ⅰ、Ⅲ型胶原和糜酶mRNA表达,Western blot检测转化生长因子β1(TGF-β1)和过氧化酶体增殖物激活受体α(PPARα)蛋白表达。结果:SHR组和糜酶抑制剂(CHI)组Ⅰ、Ⅲ型胶原mRNA、TGF-β1蛋白高于对照组,PPARα蛋白低于对照组(P<0.01,P<0.05)。SHR组糜酶mRNA高于对照组(P<0.01)。结论:糜酶参与心肌纤维化,其机制与TGF-β1蛋白上调、PPARα蛋白下调有关。
Objective:To evaluate the effect of chymase on myocardial fibrosis in spontaneously hypertensive rats(SHR).Methods: The mRNA expressions of Ⅰ-and Ⅲ-type collagen and chymase in myocardial tissue were detected by RT-PCR,and the protein expressions of transforming growth factor-β1(TGF-β1) and peroxisome proliferator-activated receptor α(PPARα) were determined by Western blot.Results: The mRNA expressions of Ⅰ-and Ⅲ-type collagen,the protein expression of TGF-β1 in SHR group and chymase inhibitor(CHI) group were higher than those in control group,but the protein expression of PPARα was lower than that in control group(P<0.01,P<0.05).The mRNA expression of chymase in SHR group was markedly higher than that in control group(P<0.01).Conclusion: Chymase is involved in myocardial fibrosis,which may be associated with the upregulation of TGF-β1 protein and the downregulation of PPARα protein.
