摘要
目的:运用神经电生理及免疫组化方法,从细胞分子生物学等不同层面探讨针药结合对慢性酒精中毒型周围神经病的治疗作用机制,为针药结合治疗慢性酒精中毒性周围神经病的临床推广提供新的理论依据。方法:成年雄性W istar大鼠50只,随机分成空白组、慢性酒精中毒模型组、弥可保组、温针组及温针加弥可保组(针药组)5组,每组各10只,治疗8周后取材并运用神经电生理检测坐骨神经传导速度,酶联免疫法检测大鼠坐骨神经组织中神经生长因子的表达。结果:模型组坐骨神经传导速度较慢,治疗后神经传导速度均加快,针药组最明显;正常坐骨神经的神经生长因子表达量极低,酒精中毒损伤后坐骨神经神经生长因子表达增高;治疗组的神经生长因子表达较模型组有不同程度升高,其中针药组升高幅度最为明显。结论:针药结合可提高酒精中毒性大鼠坐骨神经传导速度及升高酒精中毒性大鼠坐骨神经中神经生长因子的含量。
Objective:Using electrophysiology and immunohistochemistry to explore the treatment mechanism of warm needling on the alcoholic peripheral neuropathy rat model due to alcohol-free drink from different levels of cell molecular biology and to provide new theoretical basis for promotion of treatment of warm needling and herbs.Methods:50 adult male WISTAR rats weighing 200-250g were randomly divided into five groups:1: control group(10),2:chronic alcoholic model group(10),3: methycobal group(10),4:warm needling group(10),5: warm needling with methycobal group(needle plus drug group)(10).Draw materials after the 8 weeks treatment.Sciatic nerve conduction velocity was detected by electrophysiological detection.Nerve growth factor expression of sciatic nerve tissue on rats was determined by Enzyme-linked immunosorbent assay.Results:1.Sciatic nerve conduction velocity of model group was slower,and was accelerated after treatment,while that of the needle plus drug group was the most obvious one.2.The expression of sciatic nerve growth factor in normal rats is extremly low,howerver,it was increased after alcoholic injury.Compared with that of the model group,the nerve growth factor expression of the treatment group was higher in varying degrees,of which the needle plus drug group was higher than that of other groups.Conclusion:Combination of acupuncture and medicine can improve sciatic nerve conduction velocity in alcoholic rat and increase the contents of sciatic nerve growth factor in alcoholic rats.
