摘要
将带有NPT Ⅱ基因的质粒PGV用花粉管通道途径转化小麦烟农 1 0 3,共结实 1 0 2 5粒 ,其中有 71 8粒种子可以萌发出幼苗 ,在这 71 8株幼苗中 ,经卡那霉素筛选 ,得到 75棵绿色抗性植株 ,提取这些抗性植株的叶片总DNA ,作NPT Ⅱ基因的PCR扩增 ,有 37株为阳性 ,转化率为 5.1 6% ,对其中 9个PCR阳性植株的Southern杂交检测结果表明它们均为转基因植株 .
The plasmid PGV with NPT-Ⅱ gene was transferred into wheat C.V.Yannnong 103 by pollen tube pathway.1025 transformed seeds were selected with kanamycin,in which 718 germinated and 75 expressed resistence to kanamycin.Resistant green plants were determined by PCR amplificaton of transgenic fragment and confirmed by southern hybridization with a transformation efficiency of 5.16%.
