摘要
以市售腺嘌呤核苷一磷酸为原料经纯化后研制脱氧核糖核苷酸标准物质。用红外光谱法和质谱法对纯化后的腺嘌呤核苷一磷酸样品进行定性鉴定,采用高效液相色谱法对腺嘌呤核苷一磷酸样品进行定量分析。探讨了高效液相色谱法分析腺嘌呤核苷一磷酸的分离和检测条件。实验发现,有机溶剂可以显著改善腺嘌呤核苷一磷酸的出峰时间,酸度和盐离子浓度可以显著改善色谱的分离度。采用多家实验室联合分析的方法对标准物质进行定值,研制的腺嘌呤核苷一磷酸标准物质的纯度为99.61%,定值结果不确定度为0.13%(k=2)。
The reference material of adenosine-5’-phosphoric acid was characterized by mass spectrometry and infrared spectroscopy. The purity was determined by high performance liquid chromatography (HPLC), the separation and detection conditions were studied.The resolution of HPLC was influenced by the organic solvent, acidity, inorganic salts and their concentration.
