摘要
[目的]考察大孔树脂对山茶甙的纯化效果,优化其工艺条件。[方法]以吸附率、吸附量和解吸率为依据,筛选最佳大孔树脂。采用正交设计,以转移率为指标,考察pH值、吸附时间、供试液浓度、树脂与供试液体积比等因素对大孔树脂吸附的影响。[结果]YWD01G2为最佳吸附树脂。当pH6~7、时间8h、供试液浓度0.05g/mL、体积比1∶1.5时,大孔树脂对于山茶甙的吸附效果最佳。以50%乙醇洗脱,可以获得产品山茶甙含量为82.3%。[结论]大孔树脂处理可显著提高山茶甙含量,其工艺简便可行。
[Objective] To observe the effect of tea saponin purified by macro-reticular resins,optimize the technique conditions.[Method] To screen the best macro-reticular resin in terms of adsorption rate,capacity and desorption rate.Orthogonal test was used to observe the effect of pH,time,concentration and volume ratio of resin to solution on adsorption.[Result] YWD01G2 was best resin for adsorption.Its optimum conditions were pH6~7,time 8h,concentration 0.05g/mL,ratio 1:1.5.The saponin content reached 82.3% in elution of 50% ethanol.[Conclusion]Saponin content could be significantly promoted by macro-reticular resin adsorption.The technique was brief and practicable.
