脂易消对NASH大鼠肝组织LP mRNA的调控 被引量:1

摘要 [目的]观察脂易消对实验性NASH大鼠肝组织LPmRNA影响。[方法]采用高脂饮食制备NASH大鼠模型。大鼠分低、中、高剂量脂易消治疗组、易善复胶囊阳性对照组、生理盐水阴性对照组、正常对照组。酶法检测大鼠血清中肝功能ALT、AST水平;肝组织常规HE染色,观察肝细胞脂肪变性、炎症活动程度;采用RT-PCR法检测大鼠肝脏中LPmRNA的表达水平。[结果]脂易消各治疗组大鼠血清ALT、AST水平与模型组比较有显著性差异(P<0.01,P<0.05);肝细胞炎症活动度皆有明显改善(P<0.05);肝组织LPmRNA表达明显降低(P<0.01,P<0.05);但与易善复组相比,各剂量治疗组则无显著性差异(P>0.05)。[结论]脂易消通过改善肝细胞炎症活动度,减轻肝组织损伤,同时降低肝组织LPmRNA表达水平,对高脂饮食诱发的NASH有治疗作用,其疗效与易善复相似。 [Objective]To observe Zhiyixiao effect on experimental NASH rats liver tissue LP mRNA,and discuss its mechanism.[Method]Take high-lipid diet to make NASH rats models,which were divided into low,middle and high dosage groups(1,2,3)of Zhiyixiao treatment,positive control group(4)of Yishanfu capsule,NS negative control group(5)and normal control group(6).Take enzyme method to measure rats serum liver function ALT,AST,tissue routine HE dye;observe liver cells steatosis and inflammatory action;take RT-PCR to test LP mRNA expression.[Result]Compared to model groups,the ALT,AST had marked difference,the liver cells inflammatory action was improved much,and the LP mRNA expression level was much lowered in groups1,2,3;compared to group 4,there’s no obvious difference among other groups.[Conclusion]Zhiyixiao,through improving liver cells inflammatory action,reduces liver tissue injury,meanwhile lowers LP mRNA expression level,has cure function to NASH induced by high-lipid diet,which is similar to Yishanfu;its path of treating non-alcoholic fat hepatitis may be related with its reducing LP mRNA expression level.
