摘要
[目的]测定并比较南、北五味子中原儿茶酸的含量。[方法]采用高效液相色谱法,色谱条件:色谱柱:DiamonsilTMC18(250×4.6 mm,5μm);柱温:30℃;流动相:乙腈-水-冰醋酸(10:90:1);流速:1.0mL.min-1;波长:260 nm。[结果]原儿茶酸在0.11~0.97μg内线性关系良好;平均回收率为98.8%。三批南五味子中原儿茶酸的含量分别为0.10%、0.14%、0.08%,相应炮制品中的含量为0.072%、0.080%、0.054%;北五味子中的含量分别为0.112%、0.051%、0.059%,相应炮制品中的含量0.066%、0.037%、0.026%。[结论]南、北五味子中原儿茶酸的含量存在差异,南五味子中原儿茶酸的含量高于北五味子。炮制对五味子中原儿茶酸的含量有影响。
[Objective]To determine and compare the content of protocatechuic acid in Fructus Schisandrae sphenantherae and Fructus Schisandrae chinensis.[Method] HPLC is used and the chromatographic column is DiamonsilTMC18(250×4.6 mm,5 μm),the separate temperature is 30 ℃,mobile phase is acetonitrile-water-glacial acetic acid(10∶90∶1),the flow is 1.0 mL·min-1 and the detect wavelength is 260 nm.[Result](1) Linearity of protocatechuic acid is good at 0.1076-0.9684 μg;the average recovery is 98.8% with RSD less than 2.0%.(2) The content of protocatechuic acid in Fructus Schisandrae sphenantherae is 0.105%,0.137%,0.080% and in Fructus Schisandrae chinensis is 0.112%,0.051%,0.059% respectively,higher than that in the corresponding processed product.[Conclusion] There is difference between Fructus Schisandrae sphenantherae and Fructus Schisandrae chinensis on the content of protocatechuic acid,and one of the reasons leading to the difference would be the processing.
