旋毛虫3日龄成虫cDNA文库的免疫筛选

Immunoscreening of cDNA Library of Adult Trichinella spiralis

以旋毛虫感染猪血清为抗体探针 ,对旋毛虫 3日龄成虫 c DNA文库进行免疫筛选 ,将获得的阳性克隆进行测序 ,用分子生物学软件对所得 c DNA序列进行序列分析。结果从 4× 10 5个重组噬菌体中共获得 33个阳性克隆。序列分析结果显示 :共发现新 c DNA序列 16个 ,已知 c DNA序列 4个。其中阳性克隆 Zh8,9,10 ,12 ,13,14 ,2 0 ,2 6 ,2 8,2 9,36 ,5 4 ,6 8,70编码丝氨酸蛋白酶。通过免疫学筛选直接获得了具有免疫反应原性的阳性 c DNA克隆 。

cDNA library of 3 days old adult of Trichinella spiralis was screened using the swine sera artificial infected with Trichinella spiralis and the positive clones were sequenced and analysed. From 4.5×10~5 recombinant, 33 positive clones were obtained. Sequence analysis revealed 16 new genes and 4 known genes, positive clones Zh8, 9, 10, 12, 13, 14, 20, 26, 28, 29, 36, 54, 68, 70 encoded serine protease. This will lay the foundation to further study on the Trichinella spiralis antigen genes.
