摘要
以己烯雌酚 (DES)、琥珀酸酐和牛血清白蛋白 (BSA)等为主要原料 ,采用混合酸酐反应 ,首先合成己烯雌酚半琥珀酸酯 (DES- HS) ,经质谱分析鉴定后 ,再将 DES- HS结合到 BSA上 ,并用 SDS- PAGE对 DES- HS- BSA进行测定。以DES- HS- BSA免疫家兔 6次 ,用琼脂双扩散试验测定抗体。试验结果表明 ,高分辨质谱测得合成的 DES- HS分子量为36 8;BSA- HS- DES的 SDS- PAGE图谱经相关软件分析表明 ,单个 BSA上结合有 32个 DES- HS;琼脂扩散试验结果证实 ,免疫兔血清中含有抗 DES的抗体。本试验为下一步研制 DES残留免疫检测试剂盒及抗
Diethylstilbestrol hemisuccinate(DES-HS) were synthesized and coupled to bovine serum albumin(BSA) with mixed-anhydride method,and the purified DES-HS were identified by high mass spectra(HRMS),and the BSA-HS-DES were (identified) by SDS-PAGE.BSA-HS-DES were injected to big ear rabbits 6 times in certain interval.Antibody to diethylstilbestrol were determined by agar diffusion test.The results showed that the molecular weight of synthesized DES-HS in (368.195 9),and the number of DES-HS coupled to each BSA molecule is 32.The antibody to diethylstilbestrol does exist in the immunized rabbit serum.It was demonstrated that artificial immunogen of diethylstilbestrol could be synthesized by mixed-anhydride method and the specificity of the polyclonal anti-diethylstilbestrol antibodies could be produced.
