浸麻芽孢杆菌(Bacillus macerans)β-1,3-1,4-葡聚糖酶特性及其基因克隆

The Properties of β-glucanase from Bacillus macerans and Cloning of Its Gene

对浸麻芽孢杆菌 (Bacillusmacerans)所产的β- 1,3- 1,4 -葡聚糖酶特性进行了分析。结果表明 :该酶的最适温度为5 5℃ ,在 6 0℃以下酶相对稳定。通过 PCR方法扩增和克隆了该酶基因的全长 DNA序列 ,长度为 734bp,其中开放阅读框架长 717bp,编码 2 37个氨基酸。经 Blast分析 ,该序列与已登录的浸麻芽孢杆菌和多粘芽孢杆菌 (B.polymyxa)的同源性较高 ,分别为 99%和 82 %。

The properties of the β-glucanase from Bacillus macerans were studied.The results showed that the enzyme optimal temperature was 55℃,and was relative stable below 60℃.The complete DNA of β-1,3-1,4-glucanase was cloned by PCR amplification.Its sequence is 734 bp in length and contains ORF of 714 bp encoding 237 amino acids.The nucleotide sequence is highly identical to that of B.macerans and B.polymyxa accessed in GenBank,and their homology are 99% and 82%,respectively.
