替米沙坦对小鼠抗衰老klotho蛋白表达的影响及机制

Effect and mechanisms of Telmisartan on the expression of klotho protein in the brain of aging mice

目的观测替米沙坦对小鼠脑脉络丛klotho蛋白表达的影响及机制。方法 32只昆明小鼠,随机分为四组,即正常对照组、一氧化氮合酶(NOS)抑制组(L-NAME组),替米沙坦组和L-NAME+替米沙坦组,连续处理4周。免疫组织化学方法观察klotho蛋白表达;Western blot检测klotho蛋白表达水平的变化;RT-PCR检测RhoA和ROCK mRNA表达水平。结果与对照组比较,L-NAME组RhoA和ROCK mRNA表达升高(P<0.01);klotho蛋白表达明显降低(P<0.01)。给予替米沙坦后,RhoA和ROCK mRNA表达降低(P<0.01);klotho蛋白表达明显增高(P<0.01)。结论替米沙坦可干预L-NAME所致的klotho蛋白表达下调,其机制可能与抑制Rho/ROCK信号通路活性有关。
