替米沙坦对小鼠抗衰老klotho蛋白表达的影响及机制

摘要 目的观测替米沙坦对小鼠脑脉络丛klotho蛋白表达的影响及机制。方法 32只昆明小鼠,随机分为四组,即正常对照组、一氧化氮合酶(NOS)抑制组(L-NAME组),替米沙坦组和L-NAME+替米沙坦组,连续处理4周。免疫组织化学方法观察klotho蛋白表达;Western blot检测klotho蛋白表达水平的变化;RT-PCR检测RhoA和ROCK mRNA表达水平。结果与对照组比较,L-NAME组RhoA和ROCK mRNA表达升高(P<0.01);klotho蛋白表达明显降低(P<0.01)。给予替米沙坦后,RhoA和ROCK mRNA表达降低(P<0.01);klotho蛋白表达明显增高(P<0.01)。结论替米沙坦可干预L-NAME所致的klotho蛋白表达下调,其机制可能与抑制Rho/ROCK信号通路活性有关。 Objective To investigate the effect and the mechanisms of Telmisartan on the expression of klotho protein in the brain choroid plexus in mice.Methods 32 male Kunming mice were divided randomly into four groups: the normal control group,NOS blockade(L-NAME) group,Telmisartan group and L-NAME plus Telmisartan group.All animals were administered by corresponding medicine for four weeks,respectively.The expression of klotho protein in the brain choroid plexus were measured by immunohistochemistry and Western bl...
