Actin-γ在冠心病血小板中蛋白含量及白细胞mRNA转录水平的研究

Study on Actin-γ protein content in platelet and its mRNA expression from peripheral blood leukocytes in coronary heart disease patient

摘要 目的检测Actin-γ在冠心病血小板中蛋白含量和外周血白细胞mRNA表达,探讨其在冠心病发病过程中的作用。方法纳入冠心病患者40例,健康对照组20例,运用流式细胞术检测单核细胞-血小板聚集体(MPAs)以观察血小板活化状态;应用蛋白免疫印迹(Western--blot)法检测血小板Actin-γ蛋白含量;采用实时荧光定量逆转录聚合酶链式反应(RT-PCR)方法检测外周血白细胞Actin-γmRNA表达。结果与健康对照组相比,冠心病组MPA水平(P<0.01)和血小板骨架蛋白Actin-γ含量明显升高(P<0.01),且二者因冠心病不同类型有所差异;而白细胞Actin-γmRNA表达无统计学差异(P>0.05)。结论冠心病发病过程中血小板骨架蛋白Actin-γ重组为血小板活化和变形、聚集提供了机械来源,可能在冠心病疾病过程中发挥了一定的作用。 Objective Platelet cytoskeleton protein Actin-γ was identified to be involved in the development of coronary heart disease(CHD) from our previous study.However,little is known about the mechanism responsible for its contribution to CHD development.Here,we focus on the protein level and mRNA expression of Actin-γ from the active platelet to investigate the role in the progression of CHD.Methods Forty CHD patients and twenty healthy individuals were selected.Monocyte-platelet aggregates(MPAs) were determined ...
