摘要
文献报道了从冬虫夏草(Cs)中同时检出蝙蝠蛾拟青霉(Ph)、中华被毛孢(Hs)和冬虫夏草菌(Os)的多个基因型;Cs的成熟伴有化学成分的改变和僵虫体中Hs竞争性菌落形成能力的下降。同时检验了Cs成熟过程中Os突变基因型生物量的变化。应用Southern杂交检验发现:Cs的成熟伴有Ph和2组Os基因型生物量的显著增加。Os突变基因型ITS片段的EcoRⅠ酶切位点缺失。Southern检验发现:在未成熟Cs中,AT偏倚型菌在子座中的rDNA生物量占绝对优势,在僵虫体中几乎没有;而GC偏倚型菌则相反。随着Cs的成熟,AT型菌在僵虫体中大量增加;GC型菌在子座中逐渐增加,但正向和逆向排列的GC型菌的总量远低于AT型。研究结果表明:Cs的成熟伴有Ph和Os菌的显著增加和几个Os突变基因型菌在Cs子座和僵虫体中差异表达的动态变化。这些变化可能对Cs子座的萌发和成熟至关重要,为Cs无性世代的研究提供了重要线索。
Multiple reports have detailed the simultaneous detection of Paecilomyces hepiali ( Ph) , Hir sutella sinensis (Hs) and multiple mutant genotypes of Ophiocordyceps sinensis (Os) in natural Cordyceps sinensis (Cs) a, and altered Cs chemical components and declined ability of caterpillar Hs in competitive colony formation during Cs maturation. This study examined the changes in biomass ofOs mutants during Cs maturation. Southern blotting revealed increases in the biomass of Ph and two groups of Os mutants during Cs maturat ion. There are multiple, scattered point mutations within the rDNA of the Os mutants, causing absence of an EcoRI site within Os ITS sequences. The AT biased genotypes were highly pre dominated in the premature stroma, but not expressed in the premature caterpillar; while the GC biased Os expressed oppositely. With Cs maturation, the biomass of AT biased mutants was increased dramati cally in the mature caterpillar; while the total biomass of the GC biases with the forward or inverse ori ented rDNA was increased gradually, but much less than the AT biased maturat ion associates with the dynamic alterat ions of differential fungal expressions of Ph and Os mutants in Cs stroma and caterpillar. These alterat ions are of importance in Cs germination and maturat ion, pro viding key evidence for unmasking the anamorph teleomorph connection puzzle of Cs.
