摘要
研究固相化HEK工程细胞的培养和产生rhAPC的水平。先将HEK细胞由贴壁适应为无血清悬浮培养,再用海藻酸钙固定,并在无血清培养基中悬浮培养(最高约为27mg/L)。固相细胞动态培养的表达水平高于细胞静态悬浮培养的表达水平(最高约为50mg/L)。固相细胞动态培养可实现细胞的高密度培养,得到较高的表达水平。
The cultivation of immobilized HEK cells was performed and the expression level of rhAPC was detected.The adhere cells were adapted for suspension cultivation in serum-free medium,then these cells were immobilized with calcium alginate and cultured in serum-free medium.The results showed that rhAPC expression level of the immobilized cells is higher than that of non-immobilized cells.The highest expression level is 50mg/L,the high density culture of immobilized cells can be achieved.
