摘要
铁皮石斛不仅是著名的兰科观赏花卉,而且也是名贵的中药材之一,具有很高的药用价值。野生的铁皮石斛濒临灭绝,保护和挽救工作意义重大。本文采用植物组织培养方法对其进行研究,探索和总结出可实用的方法。以MS(改良)培养基附加6-BA4.0mg/L+NAA0.5mg/L+IBA0.3mg/L+0.3%蔗糖+6.5g/L琼脂,pH值5.6,作为分化增殖培养基,30天后每株产生出健壮的丛生芽5-6个,将其丛生芽分株转接到1/2MS(改良)培养基中,10天开始生根3,0天生根率90%以上,茎高5-6cm,为炼苗移栽和应用推广提供重要的科学依据。
Dendrobium candidu is more high utility value because it is not only famous Ornamental flowers,but also is one of famous traditional chinese medicinal materials.And the wild Dendrobium candidu is on the edge of extinction,so it is important to protect and save it.This paper researched the saving method using tissue culture,trying to explore and summarize the useful method to save it.The result showed that the most suitable medium for the inducement of buds was MS+6-BA 4.0mg/L+NAA 0.5mg/L IBA0.3mg/L,add 0.3% sucrose and 6.5g/L agar,pH value was 5.6 and cultural time was 30 days,and the best medium for rooting was: 1/2MS+IBA,it will grow roots after 10 days and rooting rate would be more than 90% after 30 days,and average stem height up to 5-6cm.These data will provide important scientific basis with translating them in the seedling bed and utilization or popularization of Dendrobium candidu.
