摘要 目的:研究RNA干扰(RNAi)沉默基因FAM92A1-289对HeLa细胞增殖和凋亡的影响。方法:化学合成法合成小干扰RNA(siRNA),阳离子脂质体转染FAM92A1-289 HeLa细胞;实时定量RT-PCR检测其mRNA表达,MTT法和流式细胞仪检测转染后HeLa细胞增殖、细胞周期和凋亡变化。结果:转染合成FAM92A1-289 siRNA后,HeLa细胞中FAM92A1-289 mRNA表达下降(P<0.01),细胞增殖均明显增强(P<0.01);细胞周期分布改变,S期细胞明显增多,而G2/M期细胞减少,凋亡率无变化。结论:FAM92A1-289基因调节细胞增殖活动。 Objective To study the influence of RNA interference to repress FAM92A1-289 gene on the proliferation and apoptosis of HeLa cell.Methods siRNA were chemically synthesized and transfected into FAM92A1-289 HeLa cells with lipofectamine.The mRNA expression were detected with real-time quantitive RT-PCR.The cell cycle,apoptosis and proliferation of HeLa cell were detected with flow cytometry and MTT.Results After successfully synthesized and transfected FAM92A1-289 siRNA,the expression level of FAM92A1-289 mRNA were decreased(P0.01),the cell proliferation level was increased(P0.01),obviously.The distribution of cell cycle was changed,the number of cells in S phase was increased,but in G2/M phase was decreased,and the apoptosis rate had no change.Conclusion The FAM92A1-289 gene may play an important role in the proliferation of HeLa cell.
