雄激素和高脂饮食对雌性大鼠卵巢、垂体、肾上腺超微结构的影响

Effects of androgen and high-fat diet on ultrastructures of ovary,pituitary and adrenal in female rats

目的研究雄激素和高脂饮食对雌性SD大鼠卵巢、垂体和肾上腺超微结构的影响。方法 24只21日龄雌性SD大鼠随机分为对照组、雄激素注射组和雄激素注射+高脂饮食组。8周后,检测空腹血糖及血清胰岛素、睾酮、三酰甘油和胆固醇水平;透射电子显微镜观察卵巢、垂体、肾上腺组织超微结构。结果雄激素注射组、雄激素注射+高脂饮食组大鼠血糖、胰岛素、睾酮、三酰甘油、胆固醇水平均高于对照组;卵巢呈多囊性改变,垂体、肾上腺超微结构显示凋亡细胞增多。雄激素注射+高脂饮食组大鼠肾上腺上皮细胞的细胞质内脂滴增多。结论雄激素可诱导多囊卵巢的形成,同时可导致血糖升高;联合高脂饮食可构建较理想的大鼠多囊卵巢综合征模型。

Objective To investigate the effects of androgen and high-fat diet on ultrastructures of ovary,pituitary and adrenal in female SD rats. MethodsTwenty-four female SD rats aged 21 d were randomly divided into control group,androgen injection group and androgen injection+high-fat diet group.Eight weeks after treatment,serum concentrations of glucose,insulin,testosterone,triglyceride and cholesterol were measured,and ultrastructures of ovary,pituitary and adrenal were observed by transmission electron microscopy. ResultsThe serum concentrations of glucose,insulin,testosterone,triglyceride and cholesterol in androgen injection group and androgen injection+high-fat diet group were higher than those in control group.Polycystic changes of ovaries and increasing apoptosis in pituitaries and adrenals were observed in androgen injection group and androgen injection+high-fat diet group,and increasing lipids were found in androgen injection+high-fat diet group. ConclusionAndrogen can induce the polycystic ovaries and increase the blood glucose in female rats.Androgen combined with high-fat diet may well meet the modeling requirements for polycystic ovary syndrome in female rats.
