不同分组取样方法初步构建山葡萄核心种质的研究 被引量:4

摘要 根据SSR分析结果的聚类情况,采用3种方法构建124份山葡萄种质资源的核心种质,通过比较基因型数、等位基因、有效等位基因、Shannon多样性指数和基因多样度,证明采用逐步聚类法构建的核心种质代表性较好。在此基础上,考虑到雄花山葡萄的特异性和山葡萄品种在实际生产、科学研究上的重要性,人为补充了未入选的雄花山葡萄和山葡萄品种(共10份资源)。利用该法构建了由41份材料组成的山葡萄资源核心种质(取样比例占总样本的33.1%),其中野生种质资源33份,占核心种质的80.5%;选育品种8份,占核心种质的19.5%;从花型来看,雌花资源27份,占核心种质的65.8%,两性花和雄花资源各7份,各占核心种质的17.1%。山葡萄核心种质的基因型数、等位基因、有效等位基因、Shannon多样性指数和基因多样度的保留率分别为86.7%、97.9%、100.0%、99.0%和99.8%,表明构建的41个山葡萄样本的核心种质能在遗传多样性上很好地代表初始种质。 According to the cluster result of SSR,we used 3 methods to construct core germplasm of 124 V.amurensis germ plasm resources.Comparing the genotypes,allelic gene,effective allelic number,shannon diversity index and the genetic diversity,the results showed that the core germplasm by the method of gradual clustering methods had the best representation.In order to reflect particularity of staminiferous V.amurensis and importance of V.amurensis in productive practice and scientific research,ten of them were also merged into core germplasm.The core germplasm included 41 V.amurensis(33 Wild germplasm resources and 8 cultivar germplasm resources) germ plasm resources(33.1% of the total) was constructed by gradual clustering methods.Among them,there were 27 female flower germplasms(65.8% of the total),7 hermaphroditic flower germplasms(17.1% of the total) and 7 staminiferous flower germplasms(17.1% of the total).The retention rate of the genotypes,allelic gene,effective allelic number,shannon diversity index and the genetic diversity was 86.7%,97.9%,100.0%,99.0% and 99.8%,respectively.All above results showed that the constructed core germplasm of 41 V.amurensis was commendably to reply to evaluated V.amurensis germplasm.
