葡萄组织培养快速繁殖体系研究

Research on Tissue Culture and Rapid Propagation of Grapes

以葡萄茎尖、茎段、叶柄、根尖为外植体,对葡萄外植体的选择、愈伤诱导、茎叶分化苗、不定根诱导及试管苗的练苗移栽过程进行研究。结果表明:外植体以茎尖为最佳,茎尖愈伤诱导最适培养基:l/2MS+6-BA 1.0 mg/L+IBA 0.10 mg/L,诱导率达95%;茎叶分化以1/2MS+6-BA 1.5 mg/L+IBA 0.10 mg/L,分化率达100%;生根培养以1/2MS+IBA 1.0 mg/L为佳,生根率达90.3%。

Shoot tip,stem,pttiole,root tip of grapes were used as explants to study on the choice of explants,callus induction,stem and leaf differentiation seedlings,adventitious root induction and in vitro transfer of training seedlings,planting process.The results showed that the best explant was meristem-tip,the most suitable medium for callus induction was 1/2MS+6-BA 1.0 mg/L +IBA 0.10 mg/L,inductivity 95%.The best medium for adventitious bud induction was 1/2MS+6-BA 1.5 mg/L+IBA 0.10 mg/L,the percentage of callus differentiation 100%;The most appropriate medium for adventitious root induction was 1/2MS+IBA 1.0 mg/L,rooting rate 90.3%.
