海岛棉ERF族转录因子EREB6基因克隆及特征分析 被引量:2

摘要 以电子克隆,同源扩增和RACE技术相结合的方法从海岛棉中克隆出了一条新基因,命名为EREB6,它包含一个669 bp长的开放阅读框,预期编码222个氨基酸长的蛋白质.序列分析发现预期蛋白含有一段核定位信号序列,GFP融合蛋白瞬时表达实验证明该蛋白定位于细胞核;含有一个保守的ERF域,属于ERF族;同源分析表明该转录因子和AtERF13,Pti4和TiERF1同属于B3a小组,同源性很高,暗示它们具有相似的功能. In this study,a novel gene,designated EREB6 including an open reading frame with 669 bp,and coding a putative protein with 222 aa,is cloned from sealand cotton using in silico metods combined with PCR and RACE techniques.Sequence analysis show that it shares an ERF conserved domain with AtERF13,Pti4 and TiERF1,and it belongs to B3a group of ERF subfamily,and also indicates that the induced protein contains a nucleus localization signals,which is ditermined by subcellular localization experiment.The memebers of B3 group have the most homologous sequence with EREB6,which suggests that EREB6 maybe possess the similar function with them.
