摘要
目的研究醋酸棉酚(gossypol acetic acid,GAA)对人骨肉瘤MG-63细胞增殖的影响及其可能作用机制。方法采用MTT法检测GAA对MG-63细胞的增殖抑制作用,Hoechst 33258荧光染色观察细胞核的形态变化,流式细胞仪分析细胞凋亡相关蛋白Bcl-2与Bax的表达情况。结果 GAA对MG-63细胞具有显著的增殖抑制作用,并呈剂量、时间依赖性;Hoechst 33258荧光染色可观察到明显核固缩、染色质凝集等细胞凋亡现象;流式细胞仪检测发现细胞凋亡相关基因Bcl-2表达下调,而Bax表达则相对上调。结论 GAA能够抑制人骨肉瘤MG-63细胞的增殖,促进其凋亡,其作用机制可能是通过调节凋亡相关蛋白Bcl-2、Bax实现。
Objective To investigate the mechanism of the inhibitory action of gossypol acetic acid(GAA) on the proliferation of osteosarcoma MG-63 cells in vitro.Methods The inhibition of proliferation of GAA in MG-63 cells was examined by MTT analyzes.The morphology changes of the nucleus were observed by Hoechst 33258 staining.The expression of apoptosis-associated gene protein was detected by ?ow cytometer.Results The proliferation of MG-63 cells was significantly inhibited by GAA in a dose-dependent and time-dependent manner.Distinct morphologic changes of cell apoptosis such as karyopyknosis and chromatic agglutination were observed by Hoechst 33258 staining.The expression of Bcl-2 was down-regulated while Bax was up-regulated.Conclusion GAA could inhibit proliferation of the human osteosarcoma MG-63 cells and induce apoptosis,with the mechanism of which could be related to the regulation of Bcl-2 and Bax.
