慢性乙型肝炎患者外周血单核细胞PD-L1表达对T淋巴细胞功能的抑制作用 被引量：1

摘要 目的探究外周血单核细胞PD-L1表达对慢性乙型肝炎患者T淋巴细胞功能的影响。方法流式细胞仪检测慢性乙型肝炎患者外周血CD14+单核细胞PD-L1表达。以PD-L1单克隆抗体阻断PD-L1/PD-1通路,MTT法检测慢性乙型肝炎患者外周血单核细胞和淋巴细胞共培养体系中抗-CD3抗体诱导的淋巴细胞增殖反应。以HBcAg和抗PD-L1加入单核细胞和淋巴细胞共培养体系中,ELISA测定培养上清IFN-γ或IL-10水平。结果慢性乙型肝炎患者外周血CD14+单核细胞PD-L1表达明显上调。未加入单核细胞组淋巴细胞增殖能力明显高于加入单核细胞组,而抗PD-L1抗体阻断组淋巴细胞增殖能力恢复至未加入单核细胞组水平。经HBcAg刺激,与健康人相比,慢性乙型肝炎患者培养上清IFN-γ较低而IL-10水平升高;抗PD-L1抗体阻断后慢性乙型肝炎患者培养上清IFN-γ水平显著增高而IL-10水平降低。结论慢性乙型肝炎状态下外周单核细胞PD-L1高表达对HBV特异性免疫细胞功能有抑制作用。
