摘要
目的探讨大鼠严重烫伤延迟复苏后肺组织中c-fos的表达变化及其与肺损伤的相关性。方法雄性SD大鼠96只,随机分为延迟液体复苏组(DF,n=40)、即时液体复苏组(IF,n=48)和正常对照组(NC,n=8),建立总体表面积30%的Ⅲ度烫伤模型,分别于伤后1h、6h、12h、24h、72h和168h取材。采用组织病理学、免疫组织化学染色与图像分析技术,观察肺组织病理改变与c-fos的表达。结果伤后6~24hDF组大鼠肺组织病变严重,IF组相对较轻。c-fos的阳性表达位于肺泡上皮细胞核或细胞质,其表达强度DF组与IF组分别高于NC组,且DF组高于IF组(P<0.05)。结论 c-fos表达强度的变化与严重烫伤延迟复苏大鼠肺组织损伤相关。
Objective To explore the changes and significance of c-fos after severe burn with delayed fluid resuscitation.Methods A total of 96 male SD rats were employed as the experimental models(TBSA 30%,grade Ⅲ),and they were randomly divided into 3 groups:delayed fluid resuscitation group(DF),immediate fluid resuscitation group(IF)and normal control group(NC).The lung samples were harvested at 1,6,12,24,72 and 168 h after burn respectively.Pathological changes of lung tissue and c-fos expression were detected by using histopathology,immunohistochemistry and image analysis techniques.Results Postburn 6-24 h,the pathological changes of lung tissue were more severe in DF group than those in IF group.c-fos were much higher in DF group and IF group than those in NC group(P<0.05).The increase of c-fos was observed in nucleus and cytoplasm.Conclusions The result indicates that c-fos may contribute significantly to lung injury following severe burn with delayed fluid resuscitation in rats.
