摘要
目的通过检测大肠癌组织中ABCG2在蛋白和mRNA水平的表达量,分析该基因及蛋白的表达与大肠癌临床病理的关系。方法分别采用SYBR Green实时定量RT-PCR技术及免疫组化SP法检测35例大肠癌患者的癌组织及相应癌旁和远端正常组织中ABCG2基因的mRNA及蛋白的表达;评价其基因及蛋白的表达与临床病理参数的关系。结果大肠癌组织中ABCG2蛋白的阳性表达率显著高于正常组织(P<0.05);ABCG2 mRNA在癌组织中的表达显著高于癌旁及正常组织(P<0.05);ABCG2的表达高低与肿瘤直径有关(χ2=5.55,P=0.018);肿瘤低分化患者ABCG2 mR-NA阳性表达率明显高于肿瘤中-高分化患者;ABCG2的表达与患者性别、年龄、淋巴结转移、Duke分期及组织浸润深度无明显关系。结论大肠癌ABCG2的表达水平与患者肿瘤发生、发展及分化程度密切相关,检测其表达水平不但可以预测大肠癌患者预后而且对指导制订化疗方案有一定参考价值。
Objective To investigate the expressions of ABCG2 in colorectal cancer and its correlation with clinical pathologies.Methods The expressions of ABCG2 mRNA in 35 cases of colorectal cancer,adjacent and distal non-cancerous tissues were quantified by SYBR Green real-time RT-PCR.The expression of ABCG2 protein and relationship with clinical parameters and the differential expression of genes were detected by immunohistochemistry(SP method).Results The protein positive rate of ABCG2 in colorectal cancer was significantly higher than that in normal tissue.There were statistically significant differences among cancer tissues,adjacent and distal non-cancerous tissues,and normal tissues for positive expression of ABCG2 mRNA(P<0.05).There was significant corelationship between the expression of ABCG2 and tumor diameter(χ2=5.55,P=0.018).The mRNA expressions of ABCG2 in patients with poor differentiated tumors were much higher than those patients with well differentiated tumors.No corelationship was found between the mRNA expression of ABCG2 and sex,age,lymph node metastasis,Duke staging and depth of invasion.Conclusions The expression of ABCG2 in colorectal cancer is closely related with the developing and differentiated of the tumors.To detect the levels of the ABCG2 expression not only can predict the patients' prognosis but also has reference value to guide the development of chemotherapy.
