丹溪痛风加减方对MSU致家兔急性痛风性关节炎的防治作用

The Preventive Effect of Danxi Tongfeng Variation Formula on the Gouty Arthritis Induced by Microcrystalline Monosodium Urate(MSU) of Rabbits

目的:观察丹溪痛风加减方对微晶型尿酸钠(MSU)诱导的兔急性痛风性关节炎的防治作用,并探讨其可能作用机制。方法:采用MSU诱导家兔痛风性关节炎模型,在致炎5h后对关节液进行白细胞计数,并测定其细胞因子和炎症因子含量,同时进行病理组织学检查。结果:丹溪痛风加减方高、中剂量灌胃可显著降低兔关节液白细胞数以及肿瘤坏死因子-α(TNF-α),前列腺素E2(PGE2),IL-1β,IL-8水平,减轻关节及软组织水肿和炎细胞浸润、变性坏死。结论:丹溪痛风加减方可能通过抑制炎症细胞的趋化、激活,抑制炎症因子和细胞因子的合成与释放,从而明显改善MSU诱导的兔痛风性关节炎。

Objective:To observe the preventive effect of Danxi Tongfeng variation formula on the gouty arthritis induced by microcrystalline monosodium urate(MSU) of rabbits and its mechanism.Methods:Gouty arthritis models of rabbits were established by using MSU.The number of leukocyte in the joint fluid was counted.And the quantities of cytokines and inflammatory factors were measured.HE stainings of joint and surrounding soft tissue were performed.Results:The high and middle dose of Danxi Tongfeng variation formula can significantly reduce the leukocyte count and the levels of TNF-α,PGE2,IL-1β,IL-8 in rabbit joint fluid.Also the edema of the joints and soft tissue,inflammatory infiltration as well as degeneration necrosis were also lessened.Conclusion:Danxi Tongfeng variation formula can obviously improve the MSU-induced gouty arthritis in rabbits.Its mechanisms are probably related to inhibiting the chemotaxis and activation of inflammatory cells,the synthesis and release of the inflammatory cytokines.
