大鼠肝卵圆细胞增殖模型技术难点评价及免疫组织化学鉴定

Evaluation on technical difficulties of oval cell proliferation models in rats and immunohistochemical identification

摘要 目的介绍改良大鼠肝卵圆细胞增殖模型建模中的技术难点,对卵圆细胞进行形态学观察及免疫组织化学鉴定。方法建立改良的大鼠肝卵圆细胞增殖模型,设模型组和假手术组,观察大鼠肝组织结构变化、卵圆细胞增殖情况及OV6、CD34、E-Cadherin的免疫组织化学定位。结果假手术组大鼠肝组织内仅见少量卵圆细胞,模型组可见明显肝卵圆细胞增殖反应,肝卵圆细胞胞浆、胞膜OV6、CD34、E-Cadherin染色呈阳性。模型组大鼠肝组织OV6、CD34、E-Cadherin蛋白表达的灰度值均比假手术组高(P<0.01)。结论改良模型可获得满意的大鼠肝卵圆细胞增殖,其增殖部位由汇管区小胆管旁向肝小叶内延伸。 Objective To investigate the technical difficulties of hepatic oval cell proliferation models in rats,and observe the morphology characteristics and identification of oval cells.Methods Modified hepatic oval cell proliferation model was established,and sham operation group was included as well.The structure change of hepatic tissue,proliferation of oval cells,immunohistochemical identification of OV6,CD34,E Cadherin were performed.Results Proliferation of oval cell was obviously observed in model group,however little change in sham operation group.The expression changes of OV6,CD34,E Cadherin in hepatic oval cells were showed in the model group.Conclusions The modified proliferation model is an appropriate tool to study the hepatic oval cells,which extended from bile ducts around portal area to hepatic lobules.
