摘要
目的观察三化汤对大鼠脑缺血/再灌注(CI/R)早期脑梗死体积和脑梗死区钠离子通道表达的影响,探讨其脑保护作用及机制。方法采用线栓法制作大鼠局灶性CI/R模型,将SD大鼠随机分为假手术组、模型组及三化汤治疗组,每组再分CI/R后6 h、12 h、24 h、48 h和72 h 5个亚组,用氯化三苯四氮唑(TTC)染色观察CI/R大鼠脑梗死体积,HE染色观察梗死区组织病理学变化,反转录-聚合酶链反应(RT-PCR)测定Nav1.1 mRNA表达。结果与假手术组比较,模型组在CI/R后各时间点脑梗死体积明显增加,神经细胞正常结构明显破坏,Nav1.1 mRNA表达明显降低;与模型组比较,三化汤治疗组CI/R后各时间点脑梗死体积明显缩小,缺血区脑梗死现象明显减轻,Nav1.1mRNA表达明显增高。结论三化汤能明显减小局灶性CI/R大鼠脑梗死体积,显著逆转Nav1.1mRNA表达的下降。减少Na+内流可能是其脑保护作用的机制之一。
Objective To observe the effect of sanhua decotion on early cerebral infarct volume and expression of sodium ion channel in cerebral infarct zone in rats with cerebral ischemia reperfusion(CI/R).Methods The string inserting method was employed to set up the rats′ focal cerebral CI/R model.195 SD rats were randomly divided into the sham operation group,model group and sanhua decotion treatment group.After experiment,each group was then divided into 6 hours,12 hours,24 hours,48 hours and 72 hours five subgroups.The cerebral infarct volume of CI/R rats was estimated by TTC staining,the histopathological changes in infarction zone were examined by HE staining,the expression of Nav1.1 mRNA was determined by RT-PCR.Results Compared with the sham operation group,the volume of cerebral infarction in the model group increased obviously in different time point after CI/R,the normal structure of neurons was destroyed markedly,and Nav1.1 mRNA expression was decreased clearly.Compared with the model group,the volume of cerebral infarction in the sanhua decotion treatment group was decreased obviously in different time points after CI/R,the normal structure of neurons was destroyed mildly,the phenomenon of cerebral infarction in ischemic zone were alleviated distinctly,and Nav1.1 mRNA expression was up-regulated.Conclusions Sanhua decotion could reduce the volume of cerebral infarction in rats with focal CI/R and reverse the descended expression of Nav1.1 mRNA,thereby reduce sodium ion inner flow,which may contribute to protect their brains with focal CI/R.
