前列腺素E2复合骨移植后早期排斥与愈合过程的特点 被引量:1

The characteristic of the early post-transplant rejection and the defect healing after the transplantation of prostaglandin E2 compound freeze-dried bone

摘要 目的研究前列腺素E2复合冻干骨移植后的早期排斥反应与愈合过程的特点。方法 36只大耳白兔制作大段骨缺损模型,随机分为三组(n=12):自体骨组;同种异体冻干骨组;同种异体冻干骨复合0.1 mg/L前列腺素E2组;移植后检测各组的T细胞亚群比值和骨形成、骨吸收状况。结果在移植后,前列腺素E2复合组的CD8+细胞亚群升高慢,且明显低于同种异体冻干骨组(P<0.05)。移植早期,CD4+细胞在前列腺素E2复合组有轻度的升高,与自体组和同种异体组相比差异有统计学意义(P<0.05)。血清骨钙素和血清Ⅰ型胶原交联C端肽检测结果提示,前列素E2复合组与自体组差异无统计学意义(P>0.05),与同种异体冻干骨的差异有统计学意义(P<0.05)。前列腺素E2复合冻干骨可以促进骨钙素的分泌,降低Ⅰ型胶原交联C端肽的血清浓度。结论冻干骨复合前列腺素E2后能加速移植骨与宿主骨的愈合,且能抑制冻干骨残存抗原性,促进骨形成及抑制骨吸收。 Objective To study the characteristic of the early post-transplant rejection and the defect healing after the transplantation of prostaglandin E2 compound freeze-dried bone.Methods 36 rabbits with large bone defects were randomly divided into three groups,the autogenetic bone group,the freeze-dried bone allograft group and the freeze-dried bone allograft combined with 0.1 mg/L prostaglandin E2 group.After transplantation,T-cell count,bone formation and bone reabsorption state were analyzed.Results After transplantation,compared with the freeze-dried bone allograft group,CD8+T cell count increased slowly in the prostaglandin E2 compound group.And CD4+T cell count also rised in the prostaglandin E2 compound group in first week and second week.The concentration of osteocalcin and C-telopeptides of type Ⅰ collagen as the marker of bone formation and bone absorption were significantly different between the freeze-dried bone allograft group and the prostaglandin E2 compound group(P<0.05).Conclusions Prostaglandin E2 can accelerate the healing of transplanted bone and host bone by promoting the bone formation and inhibiting the bone resorption.And prostaglandin E2 can activated CD4+ T cell to reduce the rejection.
