摘要
目的探讨骨形态发生蛋白质类(BMP)中BMP-2和BMP-7在原发和复发性人脑胶质瘤组织中的表达及意义。方法收集34例脑胶质瘤患者首次就诊及其复发后再治疗的完整临床资料和组织标本,采用免疫组织化学MaxVisionTM2法检测所有患者原发、复发的脑胶质瘤组织石蜡标本中BMP-2和BMP-7阳性细胞表达水平,并分析其与脑胶质瘤复发的关系及其影响因素。结果 BMP-2和BMP-7在原发脑胶质瘤中阳性细胞计数分别为21.33±15.87、19.82±12.89,而在复发脑胶质瘤中表达分别为33.48±17.76、34.05±15.91,复发脑胶质瘤中两者表达均显著多于原发(P<0.01)。ROC曲线显示:BMP-2和BMP-7的表达水平对胶质瘤复发的诊断的准确性为中等(曲线下面积分别为0.711和0.790,均P<0.01)。结论脑胶质瘤的复发与BMP-2和BMP-7的分泌生成增加有关,且两者表达的增高对胶质瘤复发的诊断具有重要的指导意义,这为其作为脑胶质瘤复发诊断的指标应用于临床的可行性提供了依据。
Objective To observe the expression of BMP-2 and BMP-7 in primary and recurrent human brain glioma and explore its value.Methods Complete clinical datum and the tissues of 34 glioma patients were collected.The MaxVisionTM2 method of immunohistochemistry was taken to detect the expression levels of BMP-2 or BMP-7 in the positive cells.The relationship between the expression levels of BMP-2/7 and the recurrence of glioma,as well as factors that may affect the expression levels were analyzed.Results The count of BMP-2 and BMP-7 positive cells were 21.33±15.87,19.82±12.89 in primary glioma tissues,compared to 33.48±17.76,34.05±15.91 in the recurrent glioma tissues,and the paired t test showed the difference was significant(P<0.01).Further more,receiver operating characteristic curve analysis was taken.It showed that the areas under the ROC curve for the counts of BMP-2 and BMP-7 positive cells were 0.711,0.790(P<0.01).It means that the counts of BMP-2 and BMP-7 positive cells had an important value for the diagnosis of recurrent human glioma.The accuracy was medium.Conclusions The recurrence of human brain glioma is related with the increasing secretion of BMP-2 and BMP-7 in glioma patients,and the counts of BMP-2 and BMP-7 positive cells have an important value for the diagnosis of recurrent human glioma.It provides a feasible basis for the clinic application of BMP-2 and BMP-7 to be a diagnostic indicator for recurrent brain glioma.
