芒果苷对过氧化氢诱导的晶状体氧化损伤的保护作用 被引量:5

摘要 目的研究芒果苷对过氧化氢诱导的晶状体氧化损伤的保护作用,并与维生素C进行比较。方法 MTT法检测各组药物对人晶状体上皮细胞的增殖与损伤的影响;体外培养Wistar大鼠晶状体,直接观察各组药物对晶状体混浊程度的影响;并对晶状体超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)进行测定。结果 100μmol/L的过氧化氢可以诱导晶状体上皮细胞的氧化损伤和晶状体混浊(P<0.01);12.5μmol/L、25μmol/L的芒果苷可以促进HLB3细胞的生长增殖,并可抑制过氧化氢诱导的晶状体混浊,升高SOD、GSH,降低MDA,而且其作用强于100μmol/L,200μmol/L的维生素C,差异有统计学意义(P<0.01)。结论低浓度的芒果苷可以促进晶状体上皮细胞生长,对抗过氧化氢造成的氧化损伤,且抗氧化能力强于维生素C,是有效的抗氧化剂。 Objective The experiment aimed at comparing the protective effects of mangiferin and vitamin C on the lens against hydrogen peroxide.Methods MTT test was used to detect the protective effects on HLB3 cells in different groups.The turbidity degree of cultured rat lens was observed in different groups.SOD,MDA and GSH were examined in different groups.Results Hydrogen peroxide(100 μmol/L) can affect the growth of HLB3 cells(P<0.01).Mangiferin(12.5 μmol/L,25 μmol/L) could promote the cell proliferation and preserve the lens from oxidation of hydrogen peroxide,SOD and GSH were increased and MDA was decreased by mangiferin at the concentration mentioned above,while 5 μmol/L mangiferin and 50 μmol/L vitamin C had no effect.Conclusions Mangiferin of low concentration could promote the growth of lens epithelial cell and protect the lens epithelial cells from the damage of hydrogen peroxide.Mangiferin has stronger antioxidative effect when compared with vitamin C.Maniferin may be a putative antioxidative agent in the treatment of cataract.
