miR-886-3p在肾癌组织及癌旁组织中差异表达及其临床意义 被引量:1

The differential expression and its clinic significance of miR-886-3p in renal cell carcinoma versus adjacent normal tissues

摘要 目的分析miR-886-3p在肾细胞癌与相应癌旁正常组织中的表达情况,为进一步研究miR-886-3p在肾癌发生发展中的作用奠定基础,并初步探讨其临床意义。方法收集48例肾癌手术患者癌组织和癌旁正常组织标本,分别提取组织中的总RNAs,通过逆转录获得cDNA,实时定量PCR法分析miR-886-3p在肾癌组织及癌旁正常组织中的表达情况,并分析其表达量与患者临床病理各指标之间的相关性。结果与相应癌旁正常组织对比,miR-886-3p在肾癌组织中存在显著高表达,48例标本中,有35例(72.92%)miR-886-3p表达上调,13例(27.08%)表达下调,差异有统计学意义(P=0.000),平均升高5.125倍。但miR-886-3p的表达水平与肾癌患者年龄、性别、病理类型、临床TNM分期(主要为T1N0M0和T2N0M0期)没有关系(P>0.05)。结论 miR-886-3p在肾癌组织中存在显著高表达,提示它可能与肾癌的发生与进展有关,可能成为指导肾癌诊断、治疗乃至预后判断的新的生物标志物。 Objective To investigate the expression level of miR-886-3p in 48 pairs of renal cell carcinoma(RCC) and adjacent normal tissues for further study of the role of miR-886-3p in RCC,and to explore its clinical significance.Methods The total RNAs were isolated from human kidney cancer tissues and matched adjacent normal tissues from 48 patients with RCC,reverse transcription was performed firstly to get the cDNA,the expression level of miR-886-3p in RCC and adjacent normal tissues was measured by SYBR green real-time quantitative PCR usingΔΔCT methods.Lastly,the correlation of the expression level of miR-886-3p and clinicopathological features in RCC patients,such as age,gender,pathologic type as well as clinical TNM stage,were analyzed.Results Compared with adjacent normal tissues,the expression level of miR-886-3p was up-regulated in 35/48 tissues while that was down-regulated in 13/48 tissues.Aberrant miR-886-3p expression was not correlated with age,gender,pathologic type and cTNM stage(P>0.05).Conclusions Aberrant expression of miR-886-3p could help distinguish malignant tissues from adjacent normal tissues,suggesting that aberrant miR-886-3p expression might be involved in the occurrence and development of RCC and might be used as early diagnosis markers or therapeutic targets for the patients with RCC.
