β-catenin、干细胞标志物Sox-2表达与结肠癌术后复发转移的关系 被引量:4

Expression of β-catenin and Sox-2 in colonal cancer and the relationship with metastasis

摘要 目的探讨结肠癌患者肿瘤组织中β-catenin、Sox-2表达情况及其对远处转移的预测作用。方法采用免疫组化方法检测80例结肠癌患者β-catenin、Sox-2表达情况,RT-PCR检测20例冰冻标本肿瘤组织及癌旁组织中Sox-2表达情况。结果在35例转移组患者中有23例(65.71%)β-catenin阳性表达,17例(48.57%)Sox-2阳性表达,均高于非转移组。不同的肿瘤分化程度、T分期、N分期β-catenin、Sox-2表达差异没有统计学意义。肿瘤组织中Sox-2表达明显高于癌旁组织。二者均为阳性表达转移率明显高于均为阴性表达转移率,生存分析显示不同表达状态其远处转移出现时间差异有统计学意义(P=0.0009)。转移组82.86%(29/35)的患者血清癌胚抗原(CEA)水平升高,非转移组为26.67%(12/45),两组差异有统计学意义(P=0.001)。结论血清CEA水平检测同结肠癌术后远处转移相关;组织β-catenin、Sox-2表达同结肠癌术后复发转移发生相关。 Objective To investigate the expression of β-catenin,Sox-2 in colonal carcinoma and the relationship with metastasis.MethodsImmunohistochemical analysis was used to evaluate the expression of β-catenin and Sox-2.The level of Sox-2 were evaluated by RT-PCR in tumor tissue and normal tissue adjacent to tumor.ResultsIn metastasis group,23 patients(23/35,65.71%)were with high level of β-catenin,and 17 patients(17/35,48.57%)were with high level of Sox-2.The difference was significant.Expression of β-catenin and Sox-2 were not associated with differentiation,T stage or N stage.The positive expression rate of Sox-2 in colonal carcinoma tissues was significantly higher than that of normal tissues.Metastatic rate in positive expression was higher than that in negative expression.And the survival analysis showed that time of metastasis was significant different in two groups(P=0.0009).ConclusionsThe level of CEA can predict the mestatasis of colonal cancer after operation.Both of β-catenin and Sox-2 are hopeful metastasis predictor for patients with colonal cancer.
