摘要
目的探讨蟾蜍灵对前列腺癌细胞PC3生长特性的影响及其机制。方法采用不同浓度的蟾蜍灵作用于雄激素非依赖性前列腺癌细胞PC3。四甲基偶氮唑蓝法(MTT)检测细胞增殖变化,流式细胞术检测细胞凋亡情况,Western杂交的方法检测细胞增殖和凋亡相关基因Bcl-2、Akt和P-Akt蛋白的表达。结果蟾蜍灵对前列腺癌细胞PC3的生长抑制作用呈时间和剂量依赖关系,促进细胞凋亡,并且可以抑制增殖和凋亡相关基因Bcl-2和P-Akt的表达。结论蟾蜍灵可以抑制前列腺癌细胞PC3的生长,促进细胞凋亡,通过Akt/Bcl-2途径发挥作用。
Objective To detect that if bufalin could affect the proliferation and apoptosis of human prostate cancer PC3 cell line.MethodsBufalin of different concentration was added on PC3 cells.MTT assay and flow cytometry were used to test the cell proliferation and apoptosis.Western Blot assay was used to detect the related proteins expression,such as Bcl-2,Akt,P-Akt.ResultsThe cells dealed with bufalin proliferation were inhibited(P<0.05)and apoptosis percentage increased(P<0.05)with the reacting time and bufalin dosage increased.Compared with control group,the cells of bufalin added expressed lower Bcl-2 and P-Akt protein.ConclusionsBufalin can inhibit the proliferation of PC3 cells and improve their apoptosis,which through the Bcl-2 and P-Akt signal pathway.
