摘要
目的探讨肿瘤相关的黑色素瘤抗原(MAGE-A3)在喉癌中的表达情况,及与癌生物学行为的相关性。方法用间接免疫荧光技术,从蛋白水平观察和比较MAGE-A3在喉癌、癌旁组织中的表达,分析喉癌中MAGE-A3的表达与临床病理参数的关系。结果 (1)MAGE-A3荧光绿色染色表明其主要表达于癌细胞胞质,癌组织染色呈斑状、块状、线状、絮状等,有时其中弥散着强染色的颗粒。而癌旁黏膜内仅见极弱的可疑的荧光染色颗粒。(2)癌旁黏膜中MAGE-A3蛋白表达率为5.56%(2/36)。喉癌中MAGE-A3蛋白表达率为51.92%(27/52)。MAGE-A3在癌组织中的表达与癌旁黏膜相比,差异有统计学意义(P<0.05)。(3)在关于喉癌中MAGE-A3的表达与临床病理参数的关系中,随喉癌T分级、N分级、临床分期的进展,MAGE-A3表达率均增高(P<0.05)。结论 (1)喉癌中MAGE-A3蛋白有较高表达,参与喉癌的发生和发展。(2)喉癌中MAGE-A3的表达受一些临床病理参数的影响。(3)MAGE-A3蛋白表达的差异可以对喉癌的发生、发展、转移作一定的预测,并可为喉癌靶向免疫治疗奠定理论基础。
Objective To study the expression of tumor-associated antigen MAGE-A3 (melanoma antigen-A3) protein in laryngeal carcinoma and to investigate the correlation between MAGE-A3 and the biological behaviour of carcinoma.Methods The expression of MAGE-A3 in laryngeal carcinoma as well as para-carcinoma mucosa were observed and compared by using indirect immunofluorescent technique.The relationships between expression of MAGE-A3 and clinicopathologic parameters of laryngeal carcinoma were alao analysed.Results (1)Fluorescent green staining indicated that MAGE-A3 was mainly located in the cytoplasm of cancer cells.Cancer tissues staining showed maculosus,massive,thread-like,wool etc,with some positive strong staining granules diffusing in tissue sometimes.But in para-carcinoma mucosa,there were only some exceedingly weak and suspicious fluorescent staining granules.(2)The positive expression rate of MAGE-A3 was 5.56%(2/36)in para-carcinoma mucosa.(3)The positive expression rate of MAGE-A3 was 51.92%(27/52)in laryngeal carcinoma.(4)The positive expression rate in cancer tissue was significantly higher than that of para-carcinoma mucosa(P<0.05).(5)In cancer group,clinicopathological parameters such as gender,age,smoking,differentiated degree and anatomic site had no significant differences of the positive expression of MAGE-A3,but T and N classification,stage obviously contributed to the positive expression rate of MAGE-A3(P<0.05).Conclusions (1)MAGE-A3 protein has a higher expression in laryngeal carcinoma,which contributed to the occurrence and development of cancer.(2)Expression of MAGE-A3 in laryngeal carcinoma was under the influence of some clinicopathological parameters.(3)Our data suggest that the abnormal expression of MAGE-A3 may be regarded as a parameter of prognosis about the pathogenesis,development and metastasis of laryngeal carcinoma,and that which can provide the rationale for the specific targeted immunotherapy against MAGE-A3 gene in laryngeal carcinoma.
