重组人睫状神经营养因子对在体和离体大鼠成肌细胞不同作用比较

摘要 目的:观察重组人睫状神经营养因子对在体和离体大鼠成肌细胞的生物学作用。方法:通过混合酶消化和差速贴壁分离纯化大鼠成肌细胞,用含有不同浓度的重组人睫状神经营养因子(rhCNTF)的分化液培养,观察成肌细胞增殖分化的改变情况。另外,采用胫前肌肌内注射 rhCNTF,随后利用免疫组化的方法观察在体大鼠成肌细胞的变化情况。结果:2.5~10ng/ml rhCNTF 能显著抑制亚融合成肌细胞的体外分化(P<0.01),这种抑制作用呈量效依赖性,且其作用是可逆的。rhCNTF 对在体成肌细胞前体肌卫星细胞的影响不明显,但能够使肌纤维较对照组增粗。结论:在体和离体大鼠成肌细胞对重组人睫状神经营养因子的反应性存在差异。 To observe the biological effect of rhCNTF on the rat myoblasts in vivo and in vitro. Methods:Purified rat myoblasts were obstained by means of mixed enzyme digestion and preplated methods.The proliferation and differentiation changes in the cultured rat myoblasts induced by differentiation medium(DM),in- cluding various concentrations of rhCNTF were observed.Besides,by means of immunohistochemistry method, we investigated the variation of the in vivo rat myoblasts after the injection of 0.2mg/kg rhCNTF to the rat tibialis anterior muscle.Results:2.5-10 ng/ml rhCNTF could significantly inhibit the differentiation of semi-fused rat myoblasts(P<0.01).The effect of rhCNTF was dose-dependent and also reversible.No marked effect of rhC- NTF was observed on the in vivo muscle satellite cells,except for the myofiber was thicker than that of the control group.Conclusion:In vivo and in vitro rat myoblasts have different reaction to recombinant human ciliary neurotro- phic factor.
